چکیده
Antibiotic resistance has become a global health problem for humankind. Improper use of antibiotics resulted in the increasing evolved bacterial to them. There are different types mechanisms including efflux pumps. To overcome pump activity on drugs, combinatorial therapy existing antimicrobials with natural products is promising insight prevent multidrug resistance. In this study, inhibitory action plant-derived molecule silibinin pumps Bacillus subtilis was investigated. The cellular effect investigated using minimum concentration and growth studies. addition, monitored by ethidium bromide accumulation assay organism. According results, MIC value between 100-200 µgmL-1 microplate 100 inhibited cell growth. Ethidium assays were performed at safe range (25 50 µgmL-1) eliminating death, increased concentration. results proved that significant inhibitor cells candidate eliminate mechanism.

A two-component multidrug efflux pump, EbrAB, in Bacillus subtilis.
Genes (ebrAB) responsible for ethidium resistance were cloned from chromosomal DNA of Bacillus subtilis ATCC 9372. The recombinant plasmid produced elevated resistance against ethidium bromide, acriflavine, pyronine Y, and safranin O not only in Escherichia coli but also in B. subtilis. It also caused an elevated energy-dependent efflux of ethidium in E. coli. EbrA and EbrB showed high sequence...
متن کاملD-Tyrosine as a metabolic inhibitor of Bacillus subtilis.
The d-isomer of tyrosine is a potent inhibitor of growth in transformable strain 168 of Bacillus subtilis. A d-tyrosine-resistant mutant of the inhibited strain was isolated which excreted l-tyrosine, had a diminished growth rate, and required l-phenylalanine to attain the growth rate of the wild-type parent. Mapping by deoxyribonucleate transformation located this resistance in the gene coding...
متن کاملSpread of Efflux Pump Overexpressing-Mediated Fluoroquinolone Resistance and Multidrug Resistance in Pseudomonas aeruginosa by using an Efflux Pump Inhibitor
BACKGROUND Fluoroquinolone resistance in Pseudomonas aeruginosa may be due to efflux pump overexpression and/or target mutations. We designed this study to investigate the efflux pump mediated fluoroquinolone resistance and check the increasing effectiveness of fluoroquinolones in combination with an efflux pumps inhibitor among P. aeruginosa isolates from burn wounds infections. MATERIALS AN...

while performance-based language assessment has led to an increased authenticity and content validity in the practice of writing assessment, the reliability of ratings has become a major issue. research findings have shown different reactions by native english speaker (nes) and non-native english speaker (nns) teachers to students’ writings. the focus of this study is on investigating whether i...
MOLECULAR CLONING AND EVALUATION OF WILD PROMOTER IN EXPRESSION OF BACILLUS SPHAERICUS PHENYLALANINE DEHYDROGENASE GENE IN BACILLUS SUBTILIS CELLS
To evaluate the role of wild promoter of L-phenylalanine dehydrogenase (PheDH) gene, referred to as pdh, from Bacillus sphaericus in expression, cloning of pdh gene in Bacillus subtilis was performed. The whole pdh gene was cloned in pHY300PLK shuttle vector and amplified, construct (pHYDH) then transformed in B. subtilis ISW1214 and E. coli JM109. The pdh endogenous promoter presented no effec...
